Dianthus uHTS uses Spectral Shift technology, engineered for 1536-well throughput.

Spectral Shift

Dianthus uHTS relies on Spectral Shift technology. The process is simple: label one binding partner with a fluorophore, then mix a fixed amount of it with a dilution series of the ligand. After excitation at 590 nm of the mix, the binding events are detected as shifts of spectra towards either blue or red. The detection of spectral shifts is achieved through dual-wavelength detection at 650 nm and 670 nm in an isothermal environment. This makes Dianthus uHTS precise enough to detect very subtle shifts, even when taking extremely rapid measurements. Then the Kd is calculated by plotting the ratios of the fluorescence intensities from the two wavelengths against the ligand concentration.

How does Dianthus uHTS compare to biochemical HTS assays?

Dianthus uHTS provides direct target engagement data without requiring knowledge of the binding site or functional readout. This makes it ideal for targets where biochemical or FRET-based assays are difficult to establish. You get biophysical confirmation of binding, not just activity.

What throughput can I expect?

Measure 1536 data points fully automated in less than 10 minutes. You can screen 1,000,000 compounds with robust Z' scores (>0.5, typically >0.8) in 5 days.

How much sample do I need?

As little as 7 μL per well for 1536-well plates, or 9 μL per well for 384-well plates. Target concentrations can be as low as 250 pM, minimizing sample consumption even at HTS scale.

Can Dianthus uHTS integrate with my automation system?

Yes. Dianthus uHTS integrates into existing automation systems via gRPC framework for walk-away operation and hands-free workflow processing. It can also operate as a standalone device via control software. 

What affinity range can Dianthus uHTS measure?
