Spectral Shift

Andromeda X uses highly sensitive Spectral Shift optics to selectively detect your protein of interest in crude lysate. A site-specific fluorescent label is added to the lysate containing the recombinant protein, and as a temperature gradient is introduced, the optics monitor changes in the fluorophore’s environment to detect protein unfolding.

The ratio of emitted fluorescence at 650 nm and 670 nm is used to follow protein unfolding and plot the inflection point (Ti). Compare peak heights of different expression conditions to select those that result in high expression of folded protein. Use shifts in the Ti to find conditions that stabilize your protein or to confirm ligand binding.

Andromeda X

How much sample do I need per measurement?

Andromeda X uses only 10 μL of lysate per measurement. Highly sensitive optics detect your protein of interest even at low expression levels, giving you Coomassie-level sensitivity with additional functional information in just 15 minutes.

How fast can I get data?

At the maximum heating rate of 7.0 °C/min, a full experiment with 48 samples takes less than 11 minutes. You can find the Ti values for your recombinant proteins in about 15 minutes, including labeling and measurement.

What types of expression systems can I work with?

Andromeda X works with lysates from bacterial (E. coli), mammalian (HEK293), and insect (Sf9) expression systems, as well as with cell culture supernatants for secreted proteins.

Do I need to purify my protein first?

No. Andromeda X works directly in crude lysates, so you can evaluate expression levels and thermal stability before committing to any purification scheme. This lets you eliminate unpromising constructs early and save time and resources.

What labeling options are available?

NanoTemper offers site-specific, one-step labeling kits for His-tagged (Tris-NTA), SNAP-tagged, biotinylated, and Human Fc-tagged proteins. Covalent labeling kits are also available for pre-checking binding conditions or retained protein functionality ahead of other screening methods.
